Gene targeting strategy for B-hCD48 mice.
The exon 1-4 of mouse Cd48 gene that encodes the full-length protein were replaced by human CD48 exon 1-4 in B-hCD48 mice.
Strain specific CD48 expression analysis in homozygous B-hCD48 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCD48 mice (H/H), and analyzed by flow cytometry with species-specific anti-CD48 antibody. Mouse CD48 was detectable in T cells (A), CD4+T cells (B) and CD8+T cells (C) of wild-type C57BL/6 mice. Human CD48 was exclusively detectable in T cells (A), CD4+T cells (B) and CD8+T cells (C) of homozygous B-hCD48 mice but not in wild-type C57BL/6 mice.
